蛋白质一级结构是组成蛋白质多肽链的线性氨基酸序列。蛋白测序,蛋白序列分析,蛋白一级结构分析,蛋白氨基酸序列分析是确定蛋白质全部氨基酸序列的过程。通过蛋白质测序获得的信息,有非常多有价值的用途,包括:
- 用于蛋白质的鉴定;
- 设计分子克隆的探针;
- 合成可用作免疫原的肽段等。
蛋白测序服务常见有两种主要的方法:Edman降解(Harvey和Ferrier,2011; Bauer等,1997)和质谱分析(MS)(Sahukar等,2016)。
目前使用最广的蛋白质测序和鉴定方法是质谱法,而Edman降解则是表征蛋白质N端最重要的方法之一。
